The Hormonal Drivers of Female Sexual Desire Across the Menstrual Cycle

New Insights Into How Estradiol and Progesterone Shape Sexual Motivation

Hormonal shifts across the menstrual cycle have long been suspected to influence women's sexual desire, but empirical data has often been limited or inconsistent. A study by Roney and Simmons (2013), published in Hormones and Behavior, offers some of the clearest evidence to date by closely tracking hormone levels and subjective sexual motivation in naturally cycling women.

The study provides new insight into how daily fluctuations in estradiol and progesterone influence day-to-day variations in sexual desire, suggesting a complex interplay between hormones and libido that unfolds within each menstrual cycle.

Study Overview

  • Participants: Young women with natural menstrual cycles (not on hormonal contraception).
  • Design: Daily saliva samples and daily self-reports over 1–2 cycles.
  • Hormones analyzed: Estradiol, progesterone, and testosterone.
  • Outcomes measured: Subjective sexual desire and sexual behavior.

Key Findings

1. Estradiol and Progesterone Are Primary Predictors of Daily Sexual Desire

  • Estradiol showed a positive association with sexual desire: as estradiol levels rose, so did libido.
  • Progesterone had a negative association: as progesterone increased, desire declined.
  • This dynamic was most evident around mid-cycle, when desire peaked — a pattern that aligns with the ovulatory window and fertility optimization.

2. Mid-Cycle Peak in Desire

  • A clear peak in desire around ovulation was observed.
  • The subsequent decline in desire into the luteal phase was statistically mediated by rising progesterone levels.
  • However, the initial rise in desire during the follicular phase could not be fully explained by hormone measures alone, suggesting the influence of non-hormonal factors or unmeasured hormonal signals.

3. Testosterone Not a Significant Predictor

  • Contrary to popular assumptions, testosterone did not significantly predict sexual desire when estradiol and progesterone were accounted for.
  • This challenges longstanding theories about testosterone’s role in female sexual motivation within natural cycles.

4. Sexual Behavior Trends Mirror Hormonal Patterns

  • Although not statistically robust, behavioral data showed trend-level associations in the same direction: increased sexual activity was weakly linked to higher estradiol and lower progesterone.
  • Between-cycle and between-women differences in hormone levels did not significantly predict overall differences in sexual motivation.

Implications

This study highlights estradiol and progesterone as key hormonal modulators of within-cycle sexual motivation. While sexual desire peaks near ovulation—a well-known evolutionary hypothesis—this research provides physiological evidence to support that claim. Interestingly, the study also underscores that testosterone may play a less significant role in day-to-day libido changes than previously assumed.

Conclusion

The findings from Roney and Simmons offer an important contribution to the science of female sexuality. By demonstrating that estradiol boosts, and progesterone dampens, sexual desire within the cycle, the study builds a more accurate model of how libido ebbs and flows in relation to fertility-related hormonal cues.

Future studies could benefit from incorporating additional variables—such as mood, partner availability, and relational dynamics—to better capture the multi-layered nature of female sexual motivation.
